I was a post-doctoral researcher at University of North Carolina at Chapel Hill. I just left my job to relocate at Seattle WA for family reasons, and am currently unemployed (under OPT STEM extension). I wonder in this case, am I eligible to file an application for NIW?

Yes, you can file an I-140 under EB-2 NIW while you are unemployed.  But, to accompany your petition you will need to provide a detailed, sworn statement regarding your plan to continue to work in the US in your field of expertise.
